► SAME round table: comments
  • (Cesare) microbial activity, isolated strains, identified bacteria based on 16S sequencing of bulk DNA, genes: how to join these information to understand the ecology of DHABs? We need an exact description of these ecosystems, of carbon fluxes, of mud pit methane emissions. We need a complete chemical description of the basins to build models
  • (Daniele) the most important question to address the future work is to connect microbial diversity with its function.
  • (Timmis) we set up and applied powerful techniques for the analysis of DHABs microflora, like the URA technique. We have now a complete description of the microbial diversity: we achieved one of the BIODEEP project goals.
► 2002 cruise report
  • All the participants received a CD containing all the data collected during the operations on-board.
  • The INGV sub-contract has been approved by all the participants
► WP1: 2003 cruise preparation
  • (Cesare) Modus will be on board.
  • A multicorer from Ancona will be available (lighter than the one from IMBC)
  • Video records will be improved
  • We can add new sensors to the CTD, like a turbidimeter.
► WP2: activity report and planning
  • (Gert) report on the results achieved on water/brine interface, brines and sediments of the 4 basins.
  • (John) report on the results achieved on the sediments analysis
  • (Sotiris) report on the results achieved on the sediments analysis
  • Long discussion among the participants, in order to identify which type of chemical parameters of water/brine interfaces and brines would be useful for the microbiologists work. Data will be soon available on the web-site.

WP3, WP4, WP5, WP6, WP7: activity report
  • (Eleni) report on the results achieved on the macrofauna quali/quantitative analysis in the anoxic sediments of the 4 basins, compared with the oxic sediments.
  • (Peter) report on the microscopic observation of mud pit, and on the characterization of 2 enzymes expressed by cloning the DNA of a water/brine interface enrichment culture on crude oil.
  • (Daniele) report on the characterization of the microbial diversity and abundance of eubacteria and archaea along the Urania water/brine interface, and on the methane oxidation experiment in brines and mud pit in Urania East.
  • (Sara) report on the 2001 strains collection: species identification, diversity characterization, metabolic activity screening. Report on the 2002 mixed cultures obtained
  • (Peppe) report on the 2001 strains collection: species identification, morphological typing, screening for salinity tolerance.

2nd year BIODEEP meeting
  • The next meeting will take place in Heraklion, on April 2003, 3-7.

  • (Timmis) details for the preparation of papers for general and scientific journals
  • Cesare asks to all the participant to the meeting to receive in few days all the presentations, in order to have the material useful for summarizing the work done.
► BIODEEP special issue on scientific journal
  • (Cesare): multidisciplinarity
  • (Jean-Francoise, Proteus)(Timmis) problem of patents for new strains
  • (Timmis) possibility to publish in a special issue, focused on the interactions between geosphere and biosphere.
  • (Gert) Urania Basin, from the geochemistry point of view is an extremely new environment
  • (Daniele) the microbiota of the basins, accurately sampled and precisely characterized along all the depth profile
  • (Timmis) proposal on different papers

► WP1: TUB/TFH/TECNOMARE improvements to the 2003 cruise (Hans)
  • New videocamera with zoom.
  • The problem of the first cruise with the connection between Modus and Scipack has been solved: the problem was due to the cable, which has been provided new by the manufacturer
  • Modus telemetry problems were solved after McArtney technical intervention. The new configuration has been tested at a depth of 2000 m in the Messina Strait.
  • Possibility to connect the multicorer under the Sciskid, in order to have a video-record of beach sampling >> Ancona MC?
  • Length of the cable between Modus and multicorer: minimum 10m, still to be defined.
  • Possibility to sample the mud pit and to videomonitor its surface with the new 200 m cable.
  • A new pressure sensor had been requested for the HPSS.
  • If necessary, it will be possible to mark an interesting site, e.g. the beaches, with particular signals
  • table with questions arosen at the end of the 2001 cruise and applied solutions
► PROTEUS: activity on the 2001 strains collection
  • (Jean-Francoise) Strains received from the different partners, cultivation, setting up synthetic media, the recepy of which is available for the partners. Building of libraries with L’Atalante and Urania water/brine interface, and screening for enzimatic activities.
  • (Jean-Francoise) Proteus asks to the web site administrator (Conisma Mi-Bicocca) to set up the strains database that will be published in the BIODEEP web site and will collect all the strains information as they will be obtained by Proteus or by the scientific partners.
  • (Cesare) all the correspondence between the partners and Proteus has to be sent in copy to the project coordinator
► 2003 cruises

(Cesare) 1st cruise will be for geochemists, 2nd cruise will be the next June for geologists and micro/macro biologists using Modus. Open questions:

  • which multicorer do we need?
  • Details of sampling
  • Improving the capacity of Modus to maintain the position and its mobility?
  • orientation of the camera for the record of the multicorer?
